| I. Background | |
| 腫瘤抑制素 M （Oncostatin-M ，OSM）是白細(xì)胞介素-6 (IL-6)家族的一種多效性細(xì)胞 因子，參與多種炎癥反應(yīng)，如傷口愈合、肝臟再生和骨重塑。除了 OSM，IL-6 家族成員包 括 IL-6、白血病抑制因子(LIF)、IL-11、IL-27、IL31、心營(yíng)養(yǎng)因子-1 (CT-1)、纖毛神經(jīng)營(yíng)養(yǎng) 因子(CNTF)、心營(yíng)養(yǎng)因子樣細(xì)胞因子 1 (CLCF1)。OSM 在體內(nèi)廣泛表達(dá)，T 細(xì)胞、單核/巨 噬細(xì)胞、中性粒細(xì)胞等多種免疫細(xì)胞均可表達(dá) OSM。 IL-6 受體家族的受體復(fù)合物均含有 GP130 亞基。OSM 的受體復(fù)合物均為異源二聚體， 根據(jù)受體復(fù)合物中第二亞基的不同，受體復(fù)合物可分為 I 型和 II 型兩種類型(21)。I 型 OSM 受體復(fù)合物由α亞基 GP130 和β亞基 LIFRβ (LIF 受體β亞基)組成，而 II 型 OSM 受體復(fù)合物 由α亞基 GP130 和β亞基 OSMRβ (OSM 受體β亞基)組成。當(dāng) OSM 和 OSM 受體復(fù)合物的兩 個(gè)亞基同時(shí)存在時(shí)，OSM 首先與 GP130 形成低親和力的異源二聚體，然后該異源二聚體 招募 OSMR 或 LIFR 并與之結(jié)合，并激活多種信號(hào)通路，包括 JAK/STAT、MAPK、JNK 和 PI3K/AKT 通路。 | |
